چکیده

N-cadherin cDNA was cloned from a zebrafish embryonic cDNA library. Analysis of the deduced amino acid sequence of this molecule (ZN-cadherin) revealed a high degree of homology to N-cadherins of other species, except that its pre-sequence is considerably shorter. Nevertheless, following transfection into Chinese hamster ovary (CHO) cells, the expressed protein was functionally active, namely participated in calcium-dependent intercellular interactions. Moreover, ectopic over-expression of ZN-cadherin, following mRNA microinjection into 2 4 cell embryos, caused microaggregation and uneven segregation of deep cells, resulting in distorted embryos. Developmental Northern and Western blot analyses indicated that both the mRNA and the protein first appear at gastrulation. In-situ hybridization showed that ZN-cadherin mRNA was initially present in all deep cells, and later became restricted to various epithelial and neural tissues. Whole-mount immunostaining indicated that while ZN-cadherin was already present at 50% epiboly, it became associated with cell junctions only 4-5 h later. In developing somites ZN-cadherin expression was prominent but transient. High levels of the protein were detected in epithelial somites and its expression was apparently down regulated concomitantly with the onset of myogenesis. o 1994 Wiley-Liss, Inc.

Ectopic expression and knockdown of a zebrafish sox21 reveal its role as a transcriptional repressor in early development

Sox proteins are DNA-binding proteins belonging to the HMG box superfamily and they play key roles in animal embryonic development. Zebrafish Sox21a is part of group B Sox proteins and its chicken and mouse orthologs have been described as transcriptional repressor and activator, respectively, in two different target gene contexts. Zebrafish sox21a is present as a maternal transcript in the ooc...

P-219: The Role of E-Cadherin Coding Gene (CDH1) in Pathogenesis of Endometriosis

Background: Endometriosis, a gyncological disorder, benign and common cause of infertility, is defined as the presence of endometrial glands and stroma at ectopic locations outside the uterine cavity. Clinical observations have led to the assessment that endometriosis is an invasive disease. Abnormal expression of adhesion molecules such as cadherins is likely to be an important determinant of l...

Conservation of BMP signaling in zebrafish mesoderm patterning

To investigate the conservation of mechanisms for mesodermal patterning between zebrafish and Xenopus, we isolated two cDNA clones encoding bone morphogenetic protein (BMP)-related proteins from a zebrafish cDNA library. Based on their predicted amino acid sequences, these two clones were designated as zbmp-2 and zbmp-4. Whole-mount in situ hybridization analysis revealed that in gastrula embry...

Investigation of Toxicological Effects of di-2-ethylhexyl with Fish Embryo Test (FET) in Zebrafish (Danio rerio)

Di(2-ethylhexyl)phthalate (DEHP) is a plasticizer in a variety of consumer products. Given the adverse toxicity effects of DEHP on living organisms, its high consumption and dispersal has been caused a serious concern. . Nowedays, the Fish Embryo Test (FET) method is widely used among the conventional methods of toxicology .The aim of the this study was to determine the acute toxicity of Di(2-e...

Ectopic expression of N-cadherin perturbs histogenesis in Xenopus embryos.

Xenopus embryos express N-cadherin in a pattern similar to that observed in other species, and cells expressing Xenopus N-cadherin can bind to cells expressing chicken N-cadherin in vitro. To investigate the developmental role of this molecule, we injected mRNA encoding chicken N-cadherin into one blastomere of 2-cell-stage Xenopus embryos and examined the effect of its expression on their deve...
